M220 Focused-ultrasonicator™ is designed for Next-Gen Sequencing applications requiring fragment sizes between 150bp and 5kb. Compact size and ease-of-use make the M220 Focused-ultrasonicator the ideal DNA shearing solution for MiSeq™ and PGM™ users.

The M220 runs a variety of DNA shearing protocols with Covaris SonoLab 7 software and uses both the Covaris microTUBE™ & miniTUBE™.

Integrated temperature control within the M220 eliminates the need for an external chiller, saving space and cost.

Covaris AFA (Adaptive Focused Acoustics™) transmits acoustic energy at ultrasonic frequencies (>400KHz) and very short wavelengths (~3mm) and is uniquely able to focus the energy into your sample. AFA controls the shearing forces that fragment nucleic acids to the selected size. The process is isothermal, non contact, and quiet.

Covaris AFA technology in the M220 eliminates operator induced variations, improves recoveries, increases efficiency, and provides standardized results.

The mechanical shearing method of choice
New “Single Box” Design

The M220 is designed with a small footprint for bench-top use. The focused-acoustic block has been optimized to reduce its size, and the Peltier-effect thermoelectric temperature controller is fully integrated. The notebook computer operating the SonoLab software is located on top of the instrument. As M220 operates in the ultrasonic range, very little sound is audible to the operator and so it can be comfortably used anywhere in the lab.
Focused-ultrasonicator Assembly

Each focused-ultrasonicor is calibrated by Covaris to ensure uniform performance across all instruments and allowing standardized protocols. Only 15ml of AFA-grade Water is needed to couple acoustic energy to the sample. An integrated Water Level Sensor detects when water needs to be added.
Highly Accurate Temperature Control

The SonoLab software utilizes an optimized algorithm to drive the Peltier-effect thermoelectric controller which provides precise control of the temperature regardless of the acoustic treatment. The temperature stays within +/- 1°C of the set-point during a typical 10 W average power treatment allowing true isothermal processing of your samples.
Real-Time Process Monitoring

SonoLab software gives a real time display of the parameters relevant to your acoustic treatment such as Bath Temperature, Average Power, Instrument Status, and Elapsed/ Remaining Time.
Preloaded Protocols

SonoLab comes with preloaded protocols for common applications. It includes DNA shearing protocols for 50 and 130 μl sample and for fragment size distribution ranging from 100 to 5,000 bp as well as Chromatin shearing protocols.
Integrated Quality Control

SonoLab comes with a History Tab. For every sample processed, Date and Time, User Name, AFA Settings, actual Power delivered, Bath Temperature, and customizable Sample Notes are recorded so sample tracking is easy.

The M220 Focused-ultrasonicator provides the same high-performance shearing results researchers have come to expect from Covaris S-Series & E-Series instruments. As demonstrated below, the M220 Focused-ultrasonicator reproducibly shears DNA from 150bp to 5kb in microTUBEs and miniTUBEs. 

Versatile DNA Shearing Performance

Figure above demonstrates M220 DNA shearing performance for fragment lengths from 150bp to 1.5kb in the microTUBE, and to 2, 3 and 5 kbp in the miniTUBE. The M220 provides the same versatile DNA shearing capabilities as the S220, E220, & LE220.

High reproducibility at 300bp DNA Shearing

Figure above displays the reproducibility of the M220 when shearing multiple DNA fragments to a fragment length of 300bp by the same M220. The M220 shearing performance is highly reproducible. DNA was sheared using standard Covaris microTUBEs.

Consistent 3kb Fragment Length DNA Shearing

Figure above demonstrates M220 shearing performance a fragment length of 3kb. Multiple samples were reproducibly sheared to 3kb using Covaris Blue miniTUBEs. The M220 provides consistent DNA shearing performance day-to-day and lab-to-lab.
