Principle / Procedure

NucleoMag® Tissue is designed for the rapid parallel purification of genomic DNA from animal tissues, cell cultures and bacteria in a 96-well format. After lysis of the sample genomic DNA is selectively bound to the NucleoMag® Beads. Contaminants are washed away and the DNA can be eluted from the beads under low-salt conditions. As the magnetic bead technology provides a closed system (no transfer of sample from one well to another is necessary during the preparation) the risk for cross contamination is reduced. The NucleoMag® technology can be easily adapted to common laboratory automation platforms. It can be used either with static-pin separators (e.g., NucleoMag® SEP) or magnetic separators integrated into robotic workstations. Application data

Reproducibility of DNA yield and purity

Isolation of genomic DNA using NucleoMag® Tissue from a mouse tail master lysate (resulting in 20 mg tissue per sample) on a KingFisher® instrument. The results show the reproducibility of DNA yield and DNA purity. Average yields of 16.23 μg (CV: 4.86 %) were obtained.



Genomic DNA from eukaryotic cells

Isolation of genomic DNA from cultured eukaryotic cells using NucleoMag® Tissue. DNA was isolated from 106 (A) or 5 x 105 (B) HeLa cells. Obtained yields were 4.3 μg from 106 cells or 2.6 μg from 5 x 105 cells.

Genomic DNA from bacteria

Isolation of genomic DNA from bacteria using NucleoMag® Tissue. DNA was isolated from bacterial cell pellets representing 0.2 mL (A) or 0.1 mL (B) overnight culture of E.coli K12. Obtained yields were 1.2 μg from 0.2 mL or 0.6 μg from 0.1 mL of bacterial culture.

Genomic DNA from animal tissue

Isolation of genomic DNA from mouse ear tissue (approx. 20 mg tissue per sample) using NucleoMag® Tissue. DNA yield was 5–7 μg.
